4-甲基傘形酮葡糖苷酸培養(yǎng)基(MUG Medium)原理：胨提供碳氮源；硫酸銨、硫酸錳、硫酸鋅、硫酸鎂、氯化鈉和氯化鈣提供各種離子可維持均衡的滲透壓及利于目標(biāo)菌的生長(zhǎng)；磷酸鹽為緩沖劑；亞硫酸鈉和去氧膽酸鈉為選擇性抑菌劑，抑制革蘭氏陽(yáng)性菌。
細(xì)菌的葡萄糖醛酸苷酶在堿性條件下，作用于4-甲基傘形酮-β-D葡萄糖醛酸苷（4-Methylumbellifery-β-D-Glucuronide簡(jiǎn)稱MUG）的β糖醛酸苷鍵，使其水解，釋放的4-甲基傘形酮在366nm紫外燈下產(chǎn)生藍(lán)白色熒光。97%的大腸埃希氏桿菌、10%的沙門(mén)氏菌以及少量的志賀氏菌具有葡萄糖醛酸苷酶。
4-甲基傘形酮葡糖苷酸培養(yǎng)基(MUG Medium)配制方法：稱取本品2.33g于100ml蒸餾水或去離子水中，加熱煮沸溶解，分裝,115℃滅菌20分鐘,冷卻,備用。本品略渾濁。

Principles of 4-methyl umbelliferous ketone glucosidate medium (MUG Medium): peptone provides carbon and nitrogen sources; ammonium sulfate, manganese sulfate, zinc sulfate, magnesium sulfate, sodium chloride and calcium chloride provide various ions to maintain a balanced osmotic pressure and facilitate the growth of target bacteria; phosphate as a buffer; sodium sulfite and sodium deoxycholate as selective inhibitors To suppress gram positive bacteria.
Bacterial glucuronidase acts on the beta-glucuronide bond of 4-methylumbellifery-beta-D-Glucuronide (MUG) under alkaline conditions, hydrolyzing it and releasing 4-methylumbellifery-beta-D-Glucuronide to produce blue-white fluorescence under 366 nm ultraviolet lamp. 97% of Escherichia coli, 10% of Salmonella and a small number of Shigella have glucuronidase.
The preparation method of 4-methyl umbelliferous ketone glucosidate medium (MUG Medium): weigh 2.33g in 100 ml distilled water or deionized water, boil and dissolve, separate and pack, sterilize at 115 C for 20 minutes, cool and reserve. This product is slightly cloudy.
