1，培養(yǎng)基的厚度對(duì)抑菌圈的大小有影響，故平皿中加入培養(yǎng)基要固定，以4毫米深度為宜。制備的平板使用時(shí)應(yīng)放于35℃溫箱中30分鐘去除過多的水分，以免影響抗菌藥物的擴(kuò)散。
2，接種細(xì)菌后應(yīng)在室溫放置片刻，待菌液被培養(yǎng)基吸收后，再貼紙片。但不宜放置太久，否則在貼紙片前細(xì)菌已開始生長(zhǎng)可使抑菌圈縮小。
3，培養(yǎng)溫度為35攝氏度為宜。堆放試驗(yàn)平板不超過兩個(gè)，使其受熱均勻。
4，測(cè)量抑菌量應(yīng)仔細(xì)，精確，從生長(zhǎng)刺激帶測(cè)量。
